● 产品简介:

本产品是由6条双链DNA条带组成,适用于琼脂糖凝胶电泳中DNA条带的分析。产品中已含有1×loading buffer,可根据实验需要,直接取2-5 μl电泳,使用方便,电泳图像清晰。6条带的大小分别为100,250,500,750,1000,2000 bp。其中750 bp条带浓度约为100 ng/5 μl,显示加亮带,其余条带浓度约为50 ng/ 5μl。

● 贮存、效期及运输:

-20℃贮存;有效期3年;湿冰运输。

● 使用方法:

1. 取2-5 μl本产品加入到琼脂糖凝胶的加样孔中(每1 mm×1 mm加样孔加1 μl,如果加样孔

宽于5 mm,可以适当增加上样量)就行电泳。

2. 建议电泳条件:凝胶浓度为1.0-2.0%,凝胶长度5-7 cm,电泳电压4-10 v/cm,电泳时间15-20分钟。

3. 通过核酸染料染色后紫外灯或蓝光仪观察条带。

● 注意事项:

1. 琼脂糖的质量对DNA的电泳有很大影响,电泳时请尽量使用质量优等的琼脂糖。

2. 由于PAGE电泳的特殊性,该产品不建议用于PAGE电泳。

3. 请使用新鲜配制的电泳缓冲液和新鲜配制的凝胶进行电泳,以保证Marker良好的分离效果。

4. 该Marker适用于DNA片段大小的确定和对DNA含量的粗略定量,不适用于DNA片段含量的精确定量。

5.推荐使用RealSafe系列染料(货号:GR001,GR002,GR003,GR004)染色。如果使用Goldview(货号:GV001)或EB(货号:EB001)等染料进行染色,由于此类染料带更多的正电荷(染料移动方向与核酸泳动方向相反),随着电泳时间延长,染料向大片段聚集,凝胶会出现阴阳胶现象(染料含量高的凝胶紫外下较亮,含量低的凝胶紫外下发暗),导致小分子

量DNA显色很弱或不可见。阴阳胶可以在电泳结束后浸泡染色,将胶浸没在含EB或Goldview的电泳缓冲液中摇动染色20-25 min,小分子量DNA会显色清楚。更长时间浸泡会因DNA分子扩散而使条带弥散,小分子量DNA更易弥散。

6.紫外照射下EB易分解,使DNA条带荧光变浅。DNA受紫外光照射形成嘧啶二聚体,不利于待回收片段的下游工作,因此尽可能减少紫外照射时间。

7.蔗糖会结合DNA,降低其电泳迁移率,建议DNA样品上样使用含甘油的上样缓冲液,如6×DNA上样缓冲液(双染料)(货号:DL020)。

8.大部分核酸工具酶会结合DNA,降低其电泳迁移率,大分子量DNA尤为明显,建议此类DNA样品上样使用含SDS的上样缓冲液,如6×DNA上样缓冲液(含SDS)(货号:DL090)。
